What they're doing is reconstituting a virus from it's RNA genome so they can test it. It's a lot easier to do technically and logistically than isolate the virus in human cells, keep those cells alive through shipping, and get everything to the central lab in a usable state.
Pfizer's not making new variants here. They're not altering the genome. They're simply using the genome to make the full virus (with its supporting proteins, mainly the S-protein) so they can study it.
