Presuming that the gun used was (or at least based on) a Colt SAA or a Smith & Wesson Model 3, could the ammo caliber used by both these guns be able to pierce someone's body and hit a second person with enough force to gravely injure another like what ended up happening ?

Sure its possible. A lot depends on the caliber, bullet weight, and bullet design.
I don't know the specifics of the gun, but if it was a 45 as someone suggested with live ammo weights in the 200 plus grains on many 45 cal bullets I'd have to say yes it can. As someone pointed out the area struck on the first person will also be a factor in the penetration to another target.
