Presuming that the gun used was (or at least based on) a Colt SAA or a Smith & Wesson Model 3, could the ammo caliber used by both these guns be able to pierce someone's body and hit a second person with enough force to gravely injure another like what ended up happening ?

Yes Those old guns fire lead projectiles, not hollowpoints. Lead balls and bullets don't deform like a hollowpoint going thru soft tissue so if it didn't squarely hit bones, it may well have gone thru.
