It matters because his mother was married to Charles at the time of Harry's birth. That makes Harry "legitimate issue."
Even if 20 guys showed up and DNA tested postive for being Charles's son, they would have zero claim to the throne.
It's about legitimacy and being acknowledged/accepted by the father. Even if Harry wasn't Charles's son (but sorry, he is Charles's son) it's too late now. He is legitimate and he was accepted long ago.
