No, there's nothing in the Constitution that says that's the case. Anybody can be voted in as Speaker and it doesn't have to be a sitting member of the House.
It would have to be a coordinated effort to get Trump elected as Speaker.
RINOs voting for anybody else but Trump could also ensure Nancy Pelosi retains her position as Speaker as well.
If we do see this happen, we have even more proof we're watching a movie, that I can tell you.

During the vote itself, they can vote for anyone they like. Even if they're not a member of Congress.
But before the vote, both parties DO elect someone to be their Candidate for that vote.
