I would say yes, you can centrifuge a specimen of snot or whatever and get a little pile of a complicated organic structure which can be analyzed into a consistent structure of nucleic acids, which is called the sequence. In my opinion there is no doubt that there exists a structure which replicates consistently enough that we can say, yes, there is a distinct chemical entity called for convenience COVID 19. That said, viruses mutate constantly and "spelling out" or sequencing them is very time consuming, so relatively few patients are going to have their snot definitively analyzed even for what virus, let alone variants. This is where the "in silico" comes in. Some sample is analyzed for real and the results are extrapolated on a computer.
Serious rebuttal to Kirsh's claim. When Kirsh comes out against the vaxx, he's on our side. When he comes out for the virus, he's on their side. He strikes me as a limited hangout operation -- exposing some truths in order to preserve some deeper ones. TPTB can give up on the vaxx, but not the concept of the virus: A deadly thing that is invisible and can be claimed to cause whatever damage is needed is just too useful of a tool to lose.
