The sars-cov-2 virus was CREATED in a lab, so of course its been isolated. I think all this covid virus doesnt exist, there are no such thing as viruses, talk is disinformation to send us down the wrong rabbit holes. Talk such as this is an op to distract and dissuade. It glows.
At this moment in time, it is important that we put aside such theories and focus on the gain of function crimes and what they intended to be the ultimate endpoint. Keep our eyes on the ball, and that ball is crimes against humanity on the most massive scale imaginable. Its hard to contemplate the hubris here.
Germ Theory vs Terrain Theory is still a good debate to have. Those that say Big Pharma (Rockefeller’s) favored germ theory, for obvious reasons, make a valid points.

One of my best friends insists that viruses don’t exist, bacteria don’t exist, blood cells don’t exist, basically anything you would need a microscope to look at doesn’t exist. He’s stupid. I love him, but he’s stupid. He’s not a government agent.
Sequences can be constructed with bioinformatic algorithms and heuristics mining segments from genome databases. The databases contain information for natural, synthetic, and composed organisms. I spend a year of my PhD doing exactly this.
Are you referring to sequencing the virus? What I am guessing from what you wrote is that the viral sequencing is the "best informed guess", but it hasn't been actually isolated and sequenced ? I am very eager to understand more about this from someone who has first hand experience in this.
Right, not sequenced as in uncovered in an actual organism, just sequenced as in constructed in silica. To really sequence a strand of DNA for an organism, you need a whole sample from the thing. Something that has never been obtained for a virus. No sample = no real real sequence, but if you have some pieces of what you want to pretend come from a virus, you can use a computer to compose a sequence.
My experience was twelve years ago, so my knowledge is out of date. Plus bioinformatics was not my eventual thesis topic. You have a much better grasp on the biological aspects of everything Covid than I do. I mention it just to let folks know what can be done with computers, and that it is quite common to work with gene sequences of things that have never existed.

I would say yes, you can centrifuge a specimen of snot or whatever and get a little pile of a complicated organic structure which can be analyzed into a consistent structure of nucleic acids, which is called the sequence. You can't grow viruses in a Petri dish, they need a living host. Beyond that it's a battle of words. Whatever you think of Steve Kirsch, he has a summary of arguments here, with many links.

In my opinion there is no doubt that there exists a structure which replicates consistently enough that we can say, yes, there is a distinct chemical entity called for convenience COVID 19. That said, viruses mutate constantly and "spelling out" or sequencing them is very time consuming, so relatively few patients are going to have their snot definitively analyzed even for what virus, let alone variants. This is where the "in silico" comes in. Some sample is analyzed for real and the results are extrapolated on a computer.
Serious rebuttal to Kirsh's claim. When Kirsh comes out against the vaxx, he's on our side. When he comes out for the virus, he's on their side. He strikes me as a limited hangout operation -- exposing some truths in order to preserve some deeper ones. TPTB can give up on the vaxx, but not the concept of the virus: A deadly thing that is invisible and can be claimed to cause whatever damage is needed is just too useful of a tool to lose.

A lot of times those dumb people get seeded with misinformation. They go on repeating it not knowing truth from lies.
Or they hear a few really good points, and suddenly they give all the crazy shit a pass because of it.
